摘要Fungal collections are typically preserved as dry specimens, primarily consisting of macrofungal fruiting bodies and pathogenic fungi-infected materials. The Mycological herbarium at the National Museum of Natural Science (TNM) houses approximately 37,000 such specimens, alongside a distinct collection of living fungal strains directly linked to the dry specimen collection. Cryopreservation is a long-term preservation method for the fungal strains, significantly enhancing the utility of the fungal specimens in the museum. Our cryo-collection preserves over 4,000 fungal strains in a liquid nitrogen storage system, with duplicate copies stored in a -80℃ freezer. These strains originate from spore prints, fruiting bodies, or woody substrates of specimens deposited in TNM before drying. Once fungal strains are successfully cultured on a plate medium, they are transferred to cryotubes containing cryoprotectants. The cryotubes undergo controlled-rate freezing to -70℃ before immersion in liquid nitrogen. Prior to deposition into the liquid nitrogen system, molecular identification of fungal strains is required, and the genomic DNA of each strain is stored at -30℃. Impressively, fungal strains stored for over 30 years retain viability when thawed in a water bath at 37℃. In 2023, the National Museum of Natural Science introduced an updated collection system integrating data from dry specimens, cultures, and DNA collections. The comprehensive database enhances accessibility and usability across the museum’s fungal collections.
