| 摘要 | Microbial interaction networks inferred from sequencing data are widely used to generate ecological hypotheses, yet these predicted associations are rarely verified by experimental evidence. To bridge the gap between inferred and realized networks, we integrated co-occurrence network analysis with culture-based assays in foliar fungal endophyte communities. Foliar fungal endophytes were isolated from three gymnosperm hosts (Chamaecyparis formosensis, C. obtusa var. formosana, and Calocedrus macrolepis) in Taiwan. We compared ecological networks, inferred from co-occurrence matrices, with physiological networks constructed from in vitro paired inhibition assays. Our results showed that the ecological network was considerably sparser than the physiological network, suggesting that strong environmental filtering restricts the realization of potential interactions. Despite this sparsity, a subset of the inferred ecological edges was congruent with the physiological interactions observed in the laboratory, suggesting that network inference can successfully capture authentic biotic associations. Notably, node-level analysis revealed that ecological centrality was not solely determined by competitive ability observed in vitro. Our findings demonstrate that while network inference can successfully capture specific interactions, the assembly of the endophyte community is driven by a complex interplay of biotic potential and environmental constraints. |
