摘要 | Begonia peltatifolia H. L. Li is an endemic, endangered, and potentially ornamental species with limited distribution throughout limestone forests in Hainan, China. This study aimed to determine an efficient in vitro protocol to regenerate plantlets and contribute to the commercial propagation of an endangered species. Petioles cut in 5 × 7-mm2 segments were cultivated into in vitro plantlets on a half-strength Murashige and Skoog (1/2 MS) medium supplemented with various concentrations of plant growth regulators (PGRs). After 8 weeks, the adventitious shoots grown on 1/2 MS medium with 5 μM N6-benzyladenine (BA) and 0.5 μM indole-3-butyric acid (IBA) exhibited optimal performance. In total, 66.7% of the explants produced adventitious shoots, with a maximum average shoot number and length of 5.8 and 4.6 mm, respectively. The adventitious shoots were further subcultured on the PGR-free elongation medium for 8 weeks. The induced adventitious shoots were then transferred into a rooting medium comprising 1/2 MS medium and various IBA concentrations. After 4 weeks, rooting of 100% of the elongated shoots was achieved on 1/2 MS medium containing 1.0 μM IBA. The shoots in this medium showed the highest number (11.1) and longest length (4.6 cm) of roots. |
